Characterization of Mass-Limited Benzophenanthridine Alkaloids from California Poppy Extracts by LC-MS/ microcoilNMR

Abstract
A high sensitivity LC-MS/NMR platform is applied to the profiling and identification of benzophenanthridine alkaloids from the California Poppy. The platform combines two innovations: a post-column nanosplitter to achieve nanoelectrospray MS from 4mm LC columns, while collecting 99% of the eluant for NMR and an offline microcoil NMR approach. Doing NMR offline from LC-MS accommodates the inherent differences in MS and NMR acquisition times and sample mass requirements. 2ul samples are loaded into the NMR probe using droplet microfluidics whereby the sample is carried as a droplet in immiscible fluorocarbon fluid thus increasing sample loading efficiency.
